We are exploring physicochemical signaling and cytokine expression in bone cells that may be important in arthritis and tissue engineering. Dynamic contrast-enhanced MR imaging of bone perfusion has suggested that an increase in pressure and decrease in oxygen content may be associated with bone marrow edema. We are looking at the effects of oxygen gradients on cultured osteocytes. A second set of studies is exploring the engineering of a transplantable cartilage replacement. A stem cell population has been identified in synovium that differentiates into chondrocytes. Growth factor sequences are being defined that promote chondrocyte differentiation and matrix synthesis.
